AB The regulatory and accessory proteins of human immunodeficiency virus
type 1 (HIV-1) are produced from singly or multiply spliced mRNAs. We
have used HIV-1 specific oligonucleotide primer pairs in a polymerase
chain reaction procedure on cDNA reverse transcribed from cytoplasmic
RNA extracted from lymphocytic cell lines infected with HIV-1 228200.
The amplified products were analysed by hybridisation with spliced
junction-specific oligonucleotide probes to determine splice
donor/splice acceptor combination utilisation and the temporal
expression of different mRNA species. This work has led to the
identification of two novel splice acceptor sites which are used in the
early production of a series of distinct RNAs upon acute infection.
These RNAs have also been demonstrated in infected cells by in-situ
hybridisation with the splice junction specific probes. Work is
currently being carried out to determine the function of these RNAs.
